The knowledge of chromosome dynamics is hampered by current cytological methods that disrupt the three-dimensional (3D) architecture of the nucleus. The proposed project aims to establish the method of isolation of 3D meiocytes nuclei followed by chromosome painting in the plant model species Arabidopsis thaliana. Furthermore, the protocol will be applied to several non-model species from the Brassicaceae family to trace the behavior of the individual chromosomes during meiosis.
