Polycyclic aromatic hydrocarbons (PAHs), pesticides and other compounds belong among the most important environmental pollutants. Although many mechanisms of their toxic effects were recognized (mutagenicity, immunotoxicity), new evidences show that mech anisms of epigenetic toxicity affecting processes of cancerogenesis can be involved. Project propose development, evaluation and application of the battery of in vitro tests for detection of epigenetic mechanisms of cellular and molecular toxicity. Some of these tests are still omitted of completely missing. Following are the in vitro tests included in the proposed battery: multiparametric system for studies of oxidative stress (ROS production, GSH depletion, oxidative DNA damage, lipid peroxidation), a pplication of the methods of receptor-mediated toxicity (AhR, ER) and development of new methods (PPAR), research of inhibitors of intercellular gap juntional communications (GJIC) and development of models for studies of detoxifying enzymes (CYPIA, CYP1
